To gain insight into the molecular mechanism of PuHox52-mediated low-N adaption, we performed a transcriptome sequencing analysis (RNA-seq) using the PuHox52-OE lines and WT P. ussuriensis in roots under low-N for 3 wks, as well as roots under normal-N as controls. Selected top one internodes of in vitro propagated WT and PuHox52-OE plants, which were grown on WPM for 6 d to develop consistently, and then transferred to normal N or low-N medium for 3 wks. Roots were collected from three biological replicates respectively before total RNA extraction using the method of CTAB. Library construction and sequencing were performed by Biomarker Technologies (Beijing, China) and Bioacme Technologies (Wuhan, China).
