NPM1-ALK and FOXM1-dependent CCNB1 gene expression

Expression of CCNB1, a known FOXM1 transcriptional target, is upregulated in NPM1-ALK+ ALCL cells in a NPM1-ALK and FOXM1-dependent manner (Haque et al, 2019; Leung et al, 2001). FOXM1 binds to a consensus sequence in the CCNB1 promoter as assessed by chromatin immunoprecipitation (ChIP), and knockdown of FOXM1 abrogates CCNB1 expression in NPM1-ALK+ cells. Expression of CCNB1 and other FOXM1 targets contributes to cell growth and colony formation in NPM1-ALK cell lines (Haque et al, 2019).

CCNB1（细胞周期蛋白B1）的表达在NPM1-ALK阳性间变性大细胞淋巴瘤（ALCL）细胞中上调，这一过程依赖于NPM1-ALK和FOXM1。研究表明，FOXM1与CCNB1启动子区域中的共识序列结合，这一结合作用可通过染色质免疫沉淀（ChIP）技术进行验证。FOXM1的敲低会消除NPM1-ALK阳性细胞中的CCNB1表达。CCNB1及其它FOXM1靶基因的表达有助于NPM1-ALK细胞系中的细胞生长和集落形成（Haque等，2019；Leung等，2001）。
