In summary, our study identifies and characterizes the direct targets of Olig2 in adult oligodendrocyte lineage cells. Overall design: Sox10-CreERT2 mice were crossed with Ai14 reporter mice to generate double-transgenic offspring. Tamoxifen induction labeled oligodendrocyte lineage cells with tdTomato. Ten days post-induction, whole brains were dissociated into single-cell suspensions, and tdTomato+ cells were isolated and collected by FACS for CUT&Tag sequencing.
