Regulation of p63 expression occurs by a non-canonical version of the Hippo pathway in mammary epithelial, breast carcinoma and head and neck squamous carcinoma cells Overall design: MCF10A cells (3x105 cells per well) were seeded in 6 well plates. Next day cells were transfected with 11 nM of two different siRNAs sets against YAP and TAZ or 22 nM of control siRNA; 48 h post transfection fresh media with DMSO or 5 µM of XMU-MP1 (MST1/2 inhibitor) was added and cells were harvested 6 h later. Total RNA was isolated using TRIzol reagent (Thermofisher, 15596026) following the manufacturer's instructions
