In this study we evaluated the impact of swab, silica-gel or shock-freezing in liquid nitrogen on the Saccharina microbiota. We performed 16S Metabarcoding of the V3-V4 region with the standard Illumina protocol for metabarcoding (Q5® High-Fidelity PCR Kit, AMPure XP for PCR Purification Kit and Nextera XT DNA Library Preparation Kit), and the sequences were obtained with an Illumina MiSeq sequencer using a MiSeq v3 kit (2x300bp, paired-end).
