scRNAseq analysis in control and Goblet-Y/T-Scgb1a1CreERT mouse lung tissues.

This experiment aims to examine the global cellular response in the lung tissues of Goblet-Y/T-Scgb1a1CreERT mice. Depletion of Yap and Taz in airway secretory cells resulted in goblet cell metaplasia. To elucidate the underlying cellular response, we utilized scRNA sequencing of lung tissues. Overall design: Scgb1a1CreERT+ and Scgb1a1CreERT- mice were administered daily intraperitoneal tamoxifen injections for five consecutive days. Seven days after the final dose, the lungs were harvested and processed into single-cell suspensions. The cells were then stained with fluorescently conjugated CD45 antibodies, and CD45+ and CD45- cells were isolated and mixed in a 1:1 ratio for scRNA sequencing.
