Transcriptome analysis reveals key dysregulated transcriptional programmes in uveal melanoma

In this study, we conducted whole-transcriptome sequencing in five primary Uveal Melanoma and four adjacent tissues to uncover key dysregulated the gene-regulatory circuits in Uveal Melanoma. Overall design: The present study was performed according to the Declaration of Helsinki and approved by the Ethics Committee of the Eye Hospital of Wenzhou Medical University (ethics approval no. 2022-043-K-28-02). All patients provided written informed consent. All data were anonymously analyzed. Human UM tissues were collected from 7 patients diagnosed with UM during surgical resection in the Eye Hospital of Wenzhou Medical University. The patients did not receive preoperative treatments prior to collection. Total RNA from UM tissues was isolated using TRIzol Reagent (Invitrogen; Thermo Fisher Scientific, Inc.). The OD260/OD280 ratio was used as an indicator of RNA purity. The ratio is required to be close to 2.0 for pure RNA (acceptable range, 1.8-2.1). RNA concentration and purity was measured using the NanoDrop 2000 Spectrophotometer (Thermo Fisher Scientific, Inc.).
