Creation of streptomycin-dependent mycobacteria via recombineering

Mycobacterium smegmatis mc(2)155 and Mycobacterium tuberculosis H37Rv were made streptomycin-dependent using oligonucleotide mediated recombineering. Several spontaneously streptomycin-dependent strains of M. tuberculosis H37Rv were also discovered during this project. The streptomycin-dependent phenotype was used for selection and counterselection to recombineer other mutations into M. smegmatis.
