This dataset supports a study focused on the development and characterization of a biomaterial-based device capable of generating oxygen gradients at the micrometric scale in cell cultures. The device is designed to mimic physiological microenvironments found in vivo. The material can be microstructured using digital fabrication techniques. The spatial distribution of oxygen within the culture environment inside the device was measured using scanning electrochemical microscopy (SECM). Cells were cultured within these biomimetic oxygen gradients, and malignant cell proliferation reflected adaptation to hypoxic conditions.
The dataset includes the following characterization data:
• Laser scanning optical micrographs to determine the height profile of the inkjet-printed hydrogels
• Scanning electrochemical microscopy measurements to characterize and quantify micrometric oxygen gradients within the cell culture environment
• Quantitative analysis of optical microscopy images to assess the growth of MCF7 malignant breast epithelial cells under micrometric hypoxic gradients
• Western blot analysis of hypoxia-inducible factor 1-alpha (HIF-1α) in micrometric hypoxic conditions
• Time-course analysis of MCF7 cell growth within micrometric hypoxic gradients
