LC-MicroED: Electron diffraction tilt series acquired from solvated microcrystals at room temperature suspended within liquid cells, and relevant control dry-RT ED and cryo-XRD datasets
收藏资源简介:
This deposition contains MicroED tilt series datasets in .mrc file format (acquired at a Talos F200C TEM at 200 keV on a Direct Electron Apollo Detector), or smv file format (acquired at either a TFS Spectra 300C TEM or a Tecnai F30 TEM at 300 kev on a Ceta D detector or a TVIPS XF416 detector respectively), all at room temperature. Datasets within the "LC-ED_datasets" subdirectory were all recorded from sovlated microcrystals suspended in their mother liquor and loaded into the TEM within carbon-film liquid cells. Datasets withing the "dry-ED_datasets" subdirectory serve as relevant controls acquired from microcrystals prepared dry on TEM grids and diffracted at room temperature per conventional MicroED sample preparation methods. Also included in this archive are single-crystal X-ray diffraction data acquired at 100 K from crystals of certain of the compounds interrogated here, as well as files (CIF and FCF) relevant for the structure solution of an AVAAGA peptide structure determined from liquid cell-contained crystals at room temperature. All data at 200 keV was acquired using a TEM stage rotation rate of 1 degree/second with final frames in each image stack representing 1 second integrated exposures. All data at 300 keV was acquired using a TEM stage rotation rate of 0.5 degree/second with final frames in each image stack representating 2 second integrated exposures.
