Ectopic expression of URT1 remodels poly(A) tail profiles in Nicotiana benthamiana

3’RACE-seq experiments were conducted to determine the impact of URT1 ectopic expression on the poly(A) tail length profile of both a GFP reporter mRNA co-expressed in Nicotiana benthamiana leaves and an endogenous mRNA encoding PATHOGENESIS-RELATED PROTEIN 2 (PR2) 3’RACE-seq method was used to measure poly(A) tail sizes and analyze uridylation level. This protocol is based on the ligation of a primer at the 3’ end of RNAs, and the subsequent targeted amplification by PCR of amplicons suitable for Illumina sequencing.
