All highly and poorly permeable metastases from the same mouse brain were collected by laser capture microdissection. Total RNA from both metastatic lesions and immediate microenvironment was isolated from 5 mice bearing 231-BR metastases. As control 4 healthy mouse brains were included. We used the microarray to compare the genes differentially expressed between highly vs poorly permeable metastases. This mouse microarray gives the changes in the microenvironment. The human microarray gives the profile of tumor cells in the metastases. Female athymic NIH nu/nu mice were injected intracardiacally with the brain-tropic MDA-MB-231-BR cells. Brain metastases were established after 4 weeks. Diffusion of 3 kDa Texas red dextran was used to determine highly and poorly permeable metastases.
