As a synthesized polypeptide, AH-8 can promote collagen regeneration, but the mechanism is unknown, and other potential medical benefits are unknown. In this study, various zebrafish efficacy evaluation models were constructed to systematically evaluate the potential medical efficacy of AH-8. At the same time, the promoting effect and mechanism of AH-8 on collagen regeneration were explored from the mRNA and protein level, respectively. The results show that AH-8 can promote the expression of collagen gene and elastin gene, reduce the corresponding model of zebrafish movement distance, the expression of inflammatory factors in the body, shows that AH-8 regulate collagen, relieve nerve, relieve tingling, anti-inflammatory soothing and anti-inflammatory effect. In addition, the results of RNA-seq analysis showed that the differentially expressed genes between the model group and the treated groups were enriched in pathways related to the regulation of body aging, such as TGF signaling pathway, and the differentially expressed genes were upregulated in the treated group. Significantly divergent genes were selected and combined with previous studies, and the overall trend of qPCR, ELISA and Crispr/dCas9 gene editing results was consistent with the transcriptome sequencing results. Therefore, the AH-8 promoting collagen mechanism may be by regulating key genes on the TGF signaling pathway, thus affecting the expression of related factors.
