The study aims to identify imprinted genes and imprinting control regions in the hippocampus in mice. We selected hippocampus samples from F1 mice from a reciprocal cross between genetically divergent inbred mouse strains PWK and C57Bl6. We are currently obtaining gene expression (RNA-sequencing) in the 16 F1 mice to identify genes with monoallelic expression, which can be classified as parent-of-origin specific (imprinted) or not. We are also assaying DNA methylation patterns using MeDIP-sequencing in these tissues with the aim of identifying imprinting control regions. This specific project will add to the methylation data by providing MeDIP-sequencing estimates in 8 of the 16 individuals. The overall project aims to identify and characterize imprinted genes and imprinting control regions to improve our understanding of the mechanism of imprinting.
