Fitness profiling using the yeast deletion library to determine contribution of all non-essential genes to survival under acute exposure to lethal ethanol stress

To determine the contribution of yeast genes to acute lethal ethanol stress, we performed fitness profiling of a pooled haploid yeast deletion library. This deletion library contains 4,500 deletions of most non-essential yeast genes. Comparing the abundance of post-stress gene deletions to pre-stress gene deletions allows for the quantification of the effects of each gene deletion on survival The yeast deletion library was exposed to 2 minutes of 24.5% ethanol. A pre-stress sample was saved for comparison to a post-stress sample, which was subjected to an outgrowth period until early log phase. All experiments were performed in triplicates.
