Seasoning ingredients in a medium fat diet regulate lipid metabolism [hypothalamus]
Despite a variety of seasoning ingredients in diets, little is known about their cooperative effect on animal metabolism. We fed rats a diet containing 30 wt.% instant noodle with a 26% fat-to-energy ratio for 30 days (N-group). Compared with rats that were fed the same diet without seasonings (C-group), the N-group showed lower liver triacylglycerol levels and higher fecal cholesterol levels. To assess the mechanisms underlying this phenotype, we conducted transcriptome analyses of the hypothalamic–pituitary axis (HP), liver and white adipose tissue (WAT). Our results suggest that these ingredients may affect lipid homeostasis via the HP axis. We fed rats a diet containing 30 wt.% instant noodle with a 26% fat-to-energy ratio for 30 days (N-group). For the control group, we used rats that were fed the same diet without seasonings (C-group), and the mRNA expression in the hypothalamus was compared between the two groups.

We sought to determine how the infant gut microbiota evolve during infancy, in particular in relation to hygiene-related environmental factors, allergic and atopic disorders, and a randomized dietary intervention. Therefore, 288 healthy, exclusively breastfed infants were enrolled in a dietary intervention study from three months of age. 70 were followed up at six and twelve months to study the evolution of their gut microbiota, using 16S ribosomal RNA gene targeted amplicon sequencing.

Effects of Baobab fruit powder on gut and cardiometabolic health in obesity-Protocol for a randomised placebo-controlled trial
Despite its commercial availability, its high fibre, vitamin C and polyphenol content, there are limited scientific studies exploring the cardiometabolic effects of Baobab fruit powder (BFP) in humans. Due to its high fibre content, BFP may offer a potential intervention to reduce intestinal barrier dysfunction and therefore mitigate cardiometabolic risk. A randomized, double-blind, placebo-controlled trial will be conducted with 50 apparently healthy participants living with obesity. Participants will consume either 16 g of BFP or an isocaloric placebo daily for 45 days. The primary outcome will be intestinal permeability determined using the urinary lactulose/mannitol test. Secondary outcomes include blood biomarkers in intestinal permeability (lipopolysaccharide (LPS), intestinal fatty acid binding protein (IFABP), soluble cluster of differentiation 14 (sCD14) and LPS-binding protein (LBP)), microbiota diversity and composition and cardiometabolic risk markers including glucose levels, blood lipid profiles and blood pressure. Liver and kidney function will be monitored at baseline, after 2 weeks and following 45 days of consumption as safety outcomes. The study protocol ensures rigorous, weekly monitoring of participant compliance and tolerability, along with careful tracking of potential adverse events. Intention-to-treat analysis and mixed effects models will be employed for statistical analyses. Potential selection bias and participant dropout are addressed through thorough recruitment strategies and predefined sample size calculations. This research will contribute to the growing body of knowledge on dietary interventions in the context of cardiometabolic risk, particularly in populations at risk for developing metabolic disease.

Role of DNA methylation in the dietary restriction mediated cellular memory. Mus musculus
Dietary restriction (DR) appears to create a cellular memory, which persists even after DR is discontinued.DNA methylation seems a logical mechanism by which a cellular memory could be established.we first examined the effect of short-term DR on gene expression and DNA methylation, then determined whether the changes persisted after DR was discontinued and mice returned to ad libitum (AL) feeding.We show that DR can induce substantial changes in gene expression within one month of its implementation in various tissues, and ~15-50% of these changes in gene expression persist even when DR is discontinued. We then determined whether DR induced changes in DNA methylation in the promoter of three candidate genes (Pomc, Hsph1 and Nts1) that were correlated with the changes in the expression of these genes.
