Nanopore-based direct RNA sequencing of the Trypanosoma brucei transcriptome

Nanopore direct RNA sequencing was performed on culture-derived cells of the bloodstream and insect life cycle stages of Trypanosoma brucei. Novel isoforms of annotated transcripts as well as novel non-coding RNAs were identified. 10% of the putative lncRNAs are differentially expressed, the majority being up-regulated in bloodstream cells. In addition, differential gene expression analysis based on long reads confirmed the stage-regulated genes identified by other techniques.
