Goals: Enhance the ability of Methylorubrum extorquens PA1 to grow and thrive in formate as the carbon source through adaptive laboratory evolution (ALE). Investigate the role of Me-FDH1 in formate assimilation and its impact on the growth and metabolic adaptation of M. extorquens PA1.The Methylorubrum extorquens PA1 strain was engineered to delete all fdh genes: fdh1, fdh2, fdh3, and fdh4, and additionally contains the plasmid pCM2 carrying Me-FDH1 from Methylorubrum extorquens AM1 (W0). Implement long-term ALE experiments with the control strain W0 and the evolved strain W16 (evolved from W0 after 16 weeks of continuous transfer in formate-containing medium).The strain was cultured in MSM medium with succinate as the carbon source and was collected after 24 hours of cultivation.Reference: https://doi.org/10.1016/j.biortech.2023.130187
