To determine which loci are bound by OGDH1, we performed ChIP-seq experiment using a functional OGDH1:Flag line. We found that in the Arabidopsis genome most of the OGDH1 target loci carry histone methylation marks (e.g. H3K4me3, H3K27me3 etc.). This suggests that the function of nuclear OGDH1 is linked with histone-methylation control in Arabidopsis. Nuclei were extracted from 11-d-old OGDH1:FLAG seedlings as previously described (Louwers et al., 2009), followed by chromatin immunoprecipitation with anti-FLAG M2 magnetic beads (Sigma). DNA libraries were constructed using NEBNext Ultra II DNA Library Prep Kit for Illumina (NEB), and subsequently were sequenced on NovaSeq 6000 (Illumina).
