Pre-microRNA-derived sequences in the supraspliceosome. Homo sapiens

We report the presence of microRNA in the supraspliceosome in six different HeLa samples. After the extraction of the supraspliceosome we subjected its small RNA content to NGS. We identified 304 sequences derived from pre-miRNAs. About two thirds of them are mature miRNAs, which likely shuttled back from the cytoplasm, whereas the rest account for various defined regions of the pre-miRNA. Out of these 304 pre-miRNA-derived sequences, 95 were undetected in cellular extract, and the abundance of additional 58 strongly differed from that in cellular extract. Overall design: Examination of presence of miRNA in the supraspiceosome in six different HeLa nuclear samples and one HeLa cellular sample.
