Effect of PHRF1 depletion on gene expression in HeLa cells

The goal of this study was to understand how PHRF1 regulates gene expression in HeLa cells. By conducting RNA sequencing of 3 WT and 3 KO (PHRF1 knockout) samples, we found that DNA damage response pathways were upregulated and splicing-related pathways were downregulated in KO cells versus WT cells. Overall design: PHRF1 deletion in HeLa cells was generated through CRISPR-Cas9 mediated gene editing. Total RNA was extracted from 3 WT and 3 KO Hela cell populations and prepared for RNA sequencing. Differential gene expression analysis based on the sequencing data was conducted, comparing gene expression in PHRF1 KO cells to WT cells.
