In this project we are investigating function of Foxp3 in differentiation of regulatory T cells. Foxp3 is a multidomain transcription factor that interacts with many different proteins. These interactions are important for regulation of Foxp3 and transcription. We transfected naive T cells with several mutants of Foxp3 of either DNA binding domain or protein binding domains. Analysis of differential expression in cells with wild type Foxp3 and mutants will allow us to infere how Foxp3 regulates transcription and how different regulatory proteins influence its function. This will help to get close insight into how process of differentiation of regulatory T cells is regulated by Foxp3, which is recognized as signature transcription factor for this cell type.
