RNA-seq of FACS sorted neurons overexpressing miR-203 in wild-type mice

Purpose: The goal of this study was to assess gene expression changes in neurons overexpressing miR-203 in mouse frontal cortex. Overall design: Methods: 4-6 mice were infected in frontal cortex using bilateral sterotaxis injection with AAV2/9 either overexpressing miR-203 or scrambled miRNA. The expression of AAV2/9 was restricted to neurons using synapsin promoter. 3 or 6-weeks after infection, cortex was isolated and mononuclear suspension was made using papain digestion kit. GFP+ve neurons were sorted and RNA isolated using Clontech kit and 50bp paired-end libraries were prepared using SMARter Stranded Total RNA V4 ultra low sample prep kit (Clontech). Libraries were then multiplexed and sequenced with HiSeq 4000 instrument (Illumina).
