To characterize cellular and molecular changes associated with klf4 loss of function, we performed single-cell RNAseq on control (RNAi) and klf4 (RNAi) worms Overall design: Adult male parasites were treated with either klf4-targeting dsRNA or control dsRNA for 17 days before dissociation, then live, single isolated cells were sorted by Fluorescence-activated cell sorting (FACS) and analyzed using scRNAseq.Further data processing was carried out in Seurat (v4.3)
