Rewiring of metabolism in cancer cells induced by oncogenic KRAS involves both glucose and glutamine utilization sustaining enhanced, unrestricted growth. The development of effective anti-cancer treatments targeting metabolism may be facilitated by the identification and rational combined targeting of metabolic pathways. By applying the Illumina next-generation sequencing technology, we analyzed the transcriptome profile of KRAS-mutant A549 lung cancer and HCT116 colon cancer cell lines before and after combined treatment with the glutaminase inhibitor CB-839 and the PI3K/aldolase inhibitor NVP-BKM120. The gene expression profiles were correlated with the metabolomic profiles obtained for the two cell lines under the same treatment condition.
