Blimp1/Prdm1 is required for uterine remodeling and repair in the mouse following RU486-induced abortion

Transcriptional profiling of E6.0 wild type and Blimp1 mutant decidua, 14 hours after intraperitoneal injection with RU486 or mineral oil with was performed using RNA-seq to identify the role of Blimp1 in uterine remodelling and repair post abortion. Overall design: To induce Prdm1 gene deletion in the uterus, Prdm1 BEH/+;PRCre/+ studs (Vincent et al., 2005, Soyal et al., 2005) were crossed with female Prdm1CA/CA mice (Shapiro-Shelef et al., 2003) to generate Prdm1 BEH/CA ;PRCre females, hereinafter referred to as Blimp1 mutant. Female littermates (6–10 weeks of age) were used as the wild type. At E5.5 days post coitum, females Blimp1 mutant and wild type mice were administered 150 micrograms of RU486 in mineral oil or mineral oil alone by intraperitonal injection. The decidual swellings were collected 14 hrs later and analysed by RNA-Seq.
