Primary mesenchymal CD105+CD90+CD13- and CD105+CD90+CD13+ populations from human lung were isolated and characterized using bulk population mRNA sequencing to understand differences between these populations. Single cell suspensions from 4 normal human lungs were generated by enzymatic digestion of tissue and subsequently cryopreserved. After thawing cells, fluorescence-associated cell sorting was used to sort CD45-CD235a-CD105+CD90+CD13- and CD45-CD235a-CD105+CD90+CD13+ cells and total RNA was isolated using the PicoPure™ RNA Isolation Kit (Thermo Fisher Scientific, KIT0204). cDNA libraries were prepared using SMART-Seq v4 Ultra Low Input RNA Kit for Sequencing (Takara Bio) and Nextera XT DNA Library Preparation Kit (Illumina). A total of 8 libraries were pooled and run across two lanes of a NovaSeq 6000 S2 flow cell (Reagent Kit v1.5, 200 cycles, Illumina) on a NovaSeq 6000 System (Illumina).
