As the Omicron variant emerged in late 2021, the significant waning of wildtype (WT)-based vaccine efficacy and increased breakthrough infections have raised global concerns. In this study, memory B cells (MBCs) in a cohort of Hong Kong vaccines (n=13) were investigated after Omicron BA.1, BA.2 or BA.5 breakthrough infections. Based on single B cell antibody technology, two panels of neutralizing antibodies (NAbs) derived from memory B cells were identified for genetic, functional and structural analysis. Samples from another cohort of SARS-CoV-1 survivors (n=12) who were subsequently exposed to SARS-CoV-2 were used for single cell sequencing analysis including scRNA-seq and LIBRA-seq.Using the single B cell antibody cloning technology, single B cells were sorted (Flow cytometry data) and their BCR sequences were recovered (Sequencing data). Antibodies were expressed in vitro and used to conduct ELISA and pseudovirus-based neutralization assay for screening. The cryo-EM structures of broadly NAbs (bnAbs) were analyzed.
