To investigate the molecular mechanisms of cardiac glycosides’ neuroprotection, we profiled transcriptomes of human iPSC-neurons after 72h of treatment with increasing doses of digoxin, oleandrin, proscillaridin A or vehicle alone (0.1% DMSO) using RNA sequencing. The NPC line MGH-2046-RC1 (P301L) was derived from a female individual in her 50s with FTD carrying the autosomal dominant mutation P301L (c.C1907T NCBI NM_001123066, rs63751273), and differentiated into neuronal cells over 6-8 weeks by growth factor withdrawal. The iPSC-neurons were treated with increasing doses of digoxin, oleandrin, proscillaridin A or vehicle alone (0.1% DMSO) for 72h, followed by RNA-seq.
