ETR1 receptor mutants etr1-3 (loss-of-function) and etr1-7 (gain-of-function) were treated with either ethylene or its biosynthetic precursor, ACC (1-aminocyclopropane-1-carboxylate) for 0, 1, 4, and 24 hours. WT (Col-0), etr1-3, and etr1-7 seeds were germinated on MS media (with mesh) and grown for 5 days. Then, seedlings were transferred to new MS media. For ACC treatment, new plates contained 0.75uM ACC. For ethylene treatment, seedlings were transferred to control plates and placed into chambers where they were treated with 0.3ppm ethylene. After indicated times (0hr, 1hr, 4hr, 24hr), roots were cut from seedlings and frozen on liquid nitrogen before RNA extraction.
