Analysis of RNA-binding protein PSF- and NONO-associated transcripts in breast cancer

To examine the role of PSF and NONO in breast cancer cells, PSF- and NONO- associated transcripts were co-immunoprecipitated with PSF and NONO antibodies, respectively. Purified RNAs were analyzed with NGS. Overall design: Estrogen receptor (ER)-positive breast cancer MCF-7 were treated with 17beta-estradiol (E2) or control vehicle (ethanol), and RNA-binding protein complexes were immunoprecipitated with PSF or NONO antibody, or control IgG. NONO-RNA complexes in ER-negative MDA-MB-231 cells were immunoprecipitated with PSF or NONO antibody or control IgG.
