A recombinant virus was successfully generated by homologous recombination through co-transfection with the transfer vector and then infected with ASFV GZ201801 wild type virus. The deletion mutant was purified by successive rounds of plaque purification and limiting dilution. The purified recombinant virus was named ASFV-GZdeltaI73R, and its genome sequence was analysed by nanopore sequencing, confirming the successful deletion of I73R in the recombinant virus.
