Single cell sequencing data of islets from WT and Npc1KO mice

Npc1 is a cholesterol transporter located on lysosomal membrane, and whether it is involved in regulating the function and differentiation of à cells is unclear. To understand molecular basis of the phenotype caused by Npc1 loss in à cells, we isolated islets from 2-week-old WT and Npc1KO mice for single-cell sequencing. We compared the gene expression profiles of different subpopulation of islet cells and found many changes of islet subpopulation and biological pathway occurred in Npc1KO islets. Overall design: To understand molecular basis of the phenotype caused by Npc1 loss in à cells, we isolated islets from 2-week-old WT and Npc1KO mice, digested the islet into single-cell suspension, and performed single-cell sequencing using 10X Genomics. Our islet samples were divided into two groups, WT and Npc1KO. Every islet suspension sample were digested by the islets of 3 same genotype 2-week-old female mice.
