Whole Genome Analysis of Cladosporium allicinum and Construction of Genetic Transformation System for Silage Maize

Silage corn is an important feed and economic crop. The occurrence of silage corn diseases seriously threatens the healthy development of corn industry. Cladosporium allicinum is the main pathogen causing sooty blotch of silage maize, but its genome information has not been fully analyzed, and the genetic transformation system has not been reported. In this study, the whole genome sequencing analysis of C. allicinum DTA-10 strain isolated from silage corn were carried out for the first time by second-generation sequencing technology, and the genetic transformation system was explored, the protoplast preparation conditions were clarified, and the DTA-10 gene knockout vector was constructed based on the principle of homologous recombination. The results of whole genome sequencing showed that the genome size of C. allicinum DTA-10 was 31.27 Mb, and the GC content of the whole genome was 53.44 %. A total of 10,596 predicted genes were obtained.
