Four groups were prepared (3 rat/group): (1) SCI (AAV injection and removing 2 mm spinal cord without neural network tissue [NNT] transplantation); (2) NNT (AAV injection and NNT transplantation with MuSK overexpression); (3) Agrin (AAV injection and NNT transplantation with MuSK siRNA interference); and (4) Agrin-NNT (AAV injection and NNT transplantation). Animals were euthanized at 8 week post-surgery, and 2-mm segments of the sensorimotor cortex at the AAV injection site were collected, labeled and sequencing. we revealed that local injections of AAV9-Agrin into the sensorimotor cortex stimulate regeneration of corticospinal tact and acetylcholine release.
