Single nuclei sequencing of grafts developed 14 weeks post transplantation of human embryonic stem cell derived pancreatic progenitors alone (PP) or with rat adipose derived microvessels (PPMV) into the subcutaneous pocket of diabetic (STZ-induced) Scid-beige mice. - hESC H1 line was differentiated to stage 4 pancreatic progenitors and embedded in collagen hydrogels alone (PP) or with microvessel fragments (PPMV) freshly isolated from epididymal fat of Sprague-Dawley rats. - The PP and PPMV containing hydrogels were transplanted subcutaneously into STZ-induced diabetic Scid-beige male mice. - 14 weeks post-transplantation the grafts were retrieved from 3 mice, transplanted with independent PP differentiations and 3 mice transplanted with matching PP and independent MV isolations. - The 3 corresponding grafts were merged to generate 1 PP and 1 PPMV sample. - The two samples were subjected to nuclei isolation and single nuclei sequencing.
