To examine the role of the TGF-beta/Smad3 pathway in cardiac damage by doxorubicin, we performed comparitive RNA sequencing analysis of cardiac endothelial cells isolated from WT and Smad3 knockout mice. Overall design: Experimental male mice utilized in the study received one or four weekly tail vein injections of doxorubicin, 5 mg/kg each, starting at 7 weeks of age. Control mice were injected with saline. Cardiac endothelial cells were isolated from mice treated with doxorubicin or saline 9 weeks after the last injection. RNA sequencing analysis was performed on the isolated cells.
