Biomimetic minerals are not on par with their natural counterpart, mostly due to lack of understanding regarding the biological control that is exerted during the mineral morphogenesis. Not only the physico-chemical attachment of monomers/precursors and their assembly is important, but also the biological control over the system. In addition to specific biomolecules, mechanosensing and subsequent mechanotransduction via the cytoskeleton is thought to shape the forming biomineral. We propose using sea urchin skeletogenic cell cultures to identify the influence of the cytoskeleton on biomineralization in situ, measuring different preparations of skeletogenic cells by means of XRD/XRF mapping to obtain spatially resolved information of processes taking part in shaping the nascent mineral phase and its crystallization. These experiments will allow to answer questions at the interface of the physico-chemical understanding of biomineralization and the biological control over this process.
