We used shotgun sequencing from modern bison of known sex to simulate typical ancient DNA read profiles and mapped these to the cattle reference genome. This showed that it is feasible to determine the sex of a bison sample using shotgun sequencing by examining the ratio of reads mapping to the X chromosome, even from a divergent reference genome and as little as several thousand reads. This method allows for sex determination of bison skeletal samples and archaeological material made from bison remains.
