Pro-inflammatory macrophages impair skeletal muscle regeneration in ischemic-damaged limbs by inducing precocious differentiation of satellite cells [scRNA-seq]

To investigate the molecular mechanisms responsible for failed skeletal muscle repair in the Chronic limb threatening ischemia (CLTI) limb. We used single cell RNA sequencing (scRNA-seq) to profile the transcriptomes of the skeletal muscle specimens. Hindlimb ischemia (HLI) or sham surgery was performed on young adult C57BL/6 and BALBc mice. Hindlimb skeletal muscle tissue was collected on post-op day 1 (sham and HLI) and post-op days 3 and 7 (HLI only). Skeletal muscle was dissociated into single cells, which were analyzed by scRNA-seq.
