The aim of this study was to characterize the putative prokaryotic interactions involved in methane production during mesophilic and thermophilic conditions in a fixed-bed reactor fed with tequila vinasses. During the mesophilic phase a feeding flow strategy was applied to achieve digester stability. While, during the thermophilic phase a strategy to increase the temperature was used, keeping the feeding flow of tequila vinasses constant. Biomass samples from inoculum (U3) and biofilms (U5, U99, U105, U113, U119, and U124) were collected and processed for genomic DNA extraction. Thereafter, 16S rDNA from DNA samples were sequenced by high throughput Illumina MiSeq for bacterial and archaeal communities. Biological data obtained was analyzed to characterize abundance, diversity, and population dynamics in this methane-producing microbiome.
