In this study, we tested the effect of combination therapy of adoptive transfer of OT-1 T cells, synthetic long peptide (SLP) vaccination with OVA peptide and CD3xTRP1 bispecific antibody therapy on the effect of the immune cells in the tumor microenvironment. On day 0 we inoculated C57BL/6 mice with KPC3-TRP1 tumors and adoptively transferred OT-1 T cells on day 6, administered OVA SLP vaccination on day 8 and 15, adjuvanted with TLR7/8 ligand imiquimod and IL-2 and injected CD3xTRP1 on day 17. We sacrificed the mice on day 19, took out the tumors, processed them to single cells and isolated RNA for further analysis with Nanostring PanCancer Immune Profiling cartridges. We observed a strong increase in inflammatory cell types and pathways in the combination therapies (OT-1 + OVA + CD3xTRP1 or OVA + CD3xTRP1) compared to CD3xTRP1 monotherapy.
