Biological activated carbon (BAC) was collected from an up-flow biological filter at a drinking water treatment plant (DWTP) in Japan. Bacteria were detached from the BAC using PBS, and glass beads were inoculated with the filtered PBS to form biofilms. After seven days of incubation, the beads were used in a column assay system for enrichment over two months. Three influent substrate groups were: (1) autoclaved tap water (Tap0), (2) mineral salt medium with 300 ng/L 2-methylisoborneol (MIB) (MSM300), and (3) autoclaved tap water with 1000 ng/L MIB (Tap1000). After enrichment, DNA was extracted from the beads for full-length 16S rDNA amplicon sequencing. Comparing microbial communities across these groups allows us to identify microbes enriched by MIB addition.
