Evaluation of tofacitinib in cutaneous sarcoidosis [Olink Explore 1536]

Sarcoidosis is a multisystem inflammatory disease and no molecularly targeted therapies exist. Here we evaluate the efficacy of tofacitinib (a JAK1/3 inhibitor) in an open-label trial of 10 patients with sarcoidosis and correlate clinical improvement with molecular parameters in blood and skin. Blood samples were collected from participants at baseline and again after 6 months of therapy. Samples were compared to 11 healthy, roughly age matched controls. Blood was collected in EDTA-coated tubes and centrifuged; the buffy coat was removed and stored separately from the plasma, which was aliquoted and stored at -80ºC. Plasma was analyzed using the Olink Explore 1536 panel.
