Nuclear extracts from 293T cells overexpressing Smad3 or Snad4 were mixed with an N32 double-stranded DNA oligonucleotide library, followed by immunoprecipiation of Smad proteins using anti-Flag antibody-conjugated beads. Co-precipitated oligonucleotides were PCR-amplified. After 5 cycles of selection, concentrated oligonucleotides were analyzed by NGS.
