fter subjecting the anesthetized mice to heart perfusion with PBS to remove peripheral blood, the lymph nodes in the mammary tissue were extracted. Following this, the fat pads were isolated from 8-week-old nulliparous or other specified-stage female mice. Fat pads were minced and placed in appropriate volume (1 g/10 ml) digestion mix (RPMI 1640 with 25mM HEPES, 5% fetal bovine serum, 1% penicillin-streptomycin-glutamine, 300 U/ml type 3 collagenase (Worthington)) and digested for 2h in a 100 rpm/min shaker at 37 °C, give a good shake every 15min.
