(A) BAC recombineering primers for PCR amplification of the marker cassette. Regions of homology to the NKX2-5 open reading frame are indicated in italics. Regions that are complementary to sequences within the pEnt-Emr/Zeo marker cassette are indicated in bold. (B, C) BAC screening primers for the 5′ and 3′ recombination junctions are listed. The marker cassette reverse sequencing primer binds within the EmGFP gene, and the marker cassette forward binds within the zeosin resistance gene. Both marker cassette primers can be used to for confirmation by sequencing. All primer sequences are listed 5′ to 3′. Product sizes are listed in the right-most column.
