Synthetically mannosylated antigens induce antigen-specific humoral tolerance and prevent anti-drug antibody responses to immunogenic biologics

The purpose of this study were to test the feasibility of using pMan-antigen therapy to induce humoral tolerance. We used OVA as a model protein along with TCR transgenic OT-I and OT-II to study antigen-specific CD4+ T cell phenotypes and transcriptomic responses using RNA-seq readouts. Mice were adoptively transferred with 750,000 OT-I and OT-II cells at day 0. At day 1, mice received i.v. saline, 5 µg pMan-OVA, or equimolar OVA. Four days later, mice were sacrificed and spleens and liver and peripheral lymph nodes were pooled.Each replicate was generated from fluorescent activated cell sorted OTI and OTII cells pooled from 3 mice.
