Low Cycle Numbers Multiplex PCR: a novel strategy of constructing amplicon library for NGS

The purpose of this study is to establish an efficient and economically suitable method for constructing amplicon libraries that can be operated by ordinary researchers. This study used the mouse genome as the research object, designed primers according to its DNA sequence, and mixed multiple primers at a suitable concentration. Specific amplification was achieved using low-cycle multiplex PCR, followed by magnetic bead separation of the products twice. Subsequently, PCR was used to add sequencing adapters to both ends of the separated products to form a sequencing library that meets the sequencing requirements.
