Using single-cell RNA sequencing, we pictured a dynamic landscape of the intestinal microenvironment during RIII and regeneration. We generated transcriptomes of 22,680 single cells in the intestinal microenvironment, trying to profile the dynamics of ISCs and immune cells located in the mucous layer. This large-scale single-cell transcriptome data can be used as a valuable resource for further studying the cellular mechanism of RIII and development of potential therapy strategies. To generate a dynamic single-cell map of the intestinal microenvironment related to RIII, we employed a droplet-based scRNA-seq (10x Genomics) approach to profile the transcriptomes of single cells from the intestinal tissues at day 1 (n = 4), day 3 (n = 3), day 7 (n = 3) and day 14 (n = 3) after irradiation exposure as well as the unexposed healthy intestinal tissues (n = 4).
